Extensive research and development were done in lab to develop this novel method for cannabis sex-type determination. Commonly used sex-type tests target a region of the Y chromosome that is prone to genetic variation. Genetic variation can cause results to be non-valid, missing potential male plants. The assay developed at Green Ag Labs targets a region on the Y chromosome that is highly conserved which means less genetic variation. This results in consistent and accurate identification of male plants and fewer non-determinant results.
This novel method was developed by Green Ag Labs using next generation DNA sequencing, coupled with genome assembly and annotation. This allowed us to create a custom primer set that targets a sequence on the Y chromosome only present in male plants. As part of the method validation, we worked with multiple cannabis producers to test the assay on plants that were allowed to mature to a point where the sex-type was determined based on the presence of male or female reproductive structures. The assay was able to identify all male and female plants that were tested and none of the results were non-determinant.
